Zebrafish behavioral response to ivermectin: insights into potential neurological risk
نویسندگان
چکیده
Ivermectin is a well-known and widely used anti-parasitic drug. Recently, in vitro data suggest anti-viral efficacy of the drug, albeit at much higher concentrations than currently approved. Despite warnings by several governing bodies, (uncontrolled) human use ivermectin has significantly increased during COVID-19 epidemic. This study thus aimed to elucidate potential neurological risk particularly veterinary formulation comparison pure ivermectin. Zebrafish eggs (1hpf) larvae (4dpf) were exposed range either (IVM) or (V-IVM) for period 24 hours. Behavioral responses both treatments assessed various timepoints using pentylenetetrazol assay, light–dark assay 5-day teratogenesis protocol. In addition, dissolution rates calculated treatments. Acute 4–<5dpf was similar – transient hyperlocomotion followed general hypolocomotion (ANOVA dose effect, P < 0.01). Both IVM V-IVM-treated showed significant dose-dependent 0.0001) decreases responsiveness repeated transitions, which again more pronounced IVM. These effects maintained after hours exposure. contrast, when administered prior establishment blood brain-barrier protocol, V-IVM treatment linked severe activity decline on <5dpf. Differences cannot account these differences. conclusion, current under conditions penetration across brain-barrier.

متن کاملInsights into tuberculosis from the zebrafish model.
Mycobacterium tuberculosis (MTB) continues to plague humanity because of significant gaps in our understanding of MTB infection, including the nature of a protective versus pathological host response, why antimicrobial cure is so difficult, and the ineffectiveness of vaccination. The development of a zebrafish model, utilizing infection with the natural fish pathogen Mycobacterium marinum (Mm),...
متن کاملBiosensor Zebrafish Provide New Insights into Potential Health Effects of Environmental Estrogens
BACKGROUND Environmental estrogens alter hormone signaling in the body that can induce reproductive abnormalities in both humans and wildlife. Available testing systems for estrogens are focused on specific systems such as reproduction. Crucially, however, the potential for significant health impacts of environmental estrogen exposures on a variety of body systems may have been overlooked. OB...
متن کاملRadar breaks the fog: insights into dorsoventral patterning in zebrafish.
A mong the first major events during development of metazoans is the specification of the anteroposterior (head tail or mouth anus) and the dorsoventral (back-belly) embryonic axes. The genetic network controlling dorsoventral axis specification is best understood and remarkably, it is evolutionarily conserved from fruit f lies to mammals. Labeling of single cells or small groups of cells and t...
